The PathHunter SIRPa Signaling Bioassay consists of human cells engineered to measure drug potency and detect neutralizing antibodies. Engagement of the receptor by its ligand, or agonist antibodies results in phosphorylation of the receptor’s cytosolic tail. The SH2-domain fused to EA binds the phosphorylated receptor, forcing complementation of ED and EA, resulting in formation of an active β-gal enzyme, which hydrolyzes the substrate to generate a chemiluminescent signal. The bioassay kit contains all reagents needed for the assay including pre-qualified cryopreserved cells, detection reagents, cell plating reagent, positive control agonist, and assay plates. Biotium Viability/Cytotoxity Assay Kit for Animal Live & Dead Cell (150 assays)
The Viability/Cytotoxicity Assay Kit for Animal Live & Dead Cells provides green/red fluorescent staining of live and dead cells, respectively. The assay employs two probes: Calcein AM that detects intracellular esterase activity and stains live cells green, and Ethidium homodimer III (EthD-III) that stains cells with compromised plasma membrane integrity red. The assay principal is applicable to most eukaryotic cell types, including adherent cells and certain tissues, but not bacteria or yeast, and can be used in place of trypan blue exclusion, 51Cr release, or similar methods for determining cell viability and cytotoxicity. The kit is suitable for fluorescence microscopy, fluorescence microplate reader, or flow cytometry. EthD-III, a superior alternative to the spectrally similar Ethidium homodimer I, has a higher DNA binding affinity with ~30% brighter fluorescence.Calcein (end-product of calcein AM hydrolysis): Ex/Em: 494/517 nm (pH 8); EthD-III: Ex/Em: 530/620 nm (with DNA).
Biotium CF488A and PI Apoptosis Assay Kit
Phosphatidylserine (PS) is translocated from the inner to the outer surface of the cell for phagocytic cell recognition. The human anticoagulant, Annexin V, is a 35 kDa Ca2 -dependent phospholipid protein with a high affinity for PS. Annexin V labeled with CF488A (Abs/Em= 490/515 nm) can identify apoptotic cells in green by binding to PS exposed on the outer leaflet. Our CF488A dye is superior to fluorescein/FITC as it is not affected by pH and has much better photostability. Propidium iodide (PI) is impermeant to live cells or early apoptotic cells, but stains necrotic cells and late apoptotic cells with red fluorescence (Abs/Em with DNA = 535/617 nm). Both PI and Annexin V-CF488A can be excited using the 488 nm line of a flow cytometer. The kit has reagents sufficent for 100 assays.
